The ABI 3730xl DNA Analyzer, a 96-capillary sequencer, is the most robust sequencer on the market. With its production capacity of more than one million nucleotides per day, it is ideal for executing mega-large sequencing projects. It was brought to the market in 2002 by Applied Biosystems

The 3730xl Analyzer provides your laboratory
and production facility with very fast high quality genetic analysis and throughput. The high signal-to-noise ratio ensures you get high-quality data -- even when you use low-concentration samples and reagents. Labor-saving automation features minimize hands-on time and enable you to analyze more data more efficiently. With the 3730xl DNA Analyzer, you get the highest-quality data from a wide array of applications. Load and run up to 8 x 96 templates in continuous automated operation with less than 1 hour of instrument attendance/labor per 24 hour period.  In addition to being ideal for high-throughput sequencing, the analyzer's optimized application assays, instrument, and analysis software also provide a complete solution for genotyping and resequencing.

In addition to its 96 capillaries, the ABI 3730xl is equipped with an electrophoresis system, a laser, and a CCD camera. The capillaries, having a diameter of approximately 250 um, are filled with a polymer that acts as a molecular sieve. The DNA molecules are introduced into one end of the capillaries by electroinjection, and migrate through these capillaries under the attraction of high voltage (8500 V) at which point the molecules are separated according to their lengths. Close to the anode, a laser passes through each capillary and excites the fluorescent molecules incorporated onto the DNA during the reaction sequence. A CCD camera collects the information emitted by the fluorescent molecules as they pass in front of the laser. As each one of the four nucleic bases (A, C, G, and T) associates with a specific fluorochrome, the system distinguishes one base from another according to their respective wavelengths emitted. Subsequent data is analyzed, "base called," and filed when electrophoresis is complete.

DESCRIPTION OF FUNCTIONS AND CAPABILITIES of the ABI 3700 sequencer:

The capabilities to use multiple dyes to permit multiplexing or support dye-terminator chemistry are essential to the users of this instrument and to our productivity. 3700 is compatable with BigDye chemistry which is the state-of-the-art dye-labeled terminator chemistry.

These systems are used extensively by the genome centers, Celera, TIGR, etc. to sequence the human genome. The 3700 is a capillary electrophoresis system that can be used to sequence up to 96 DNA templates per run. The instrument accommodates up to four 96-well sample plates; a robotic arm loads the samples onto the capillaries; and electrophoresis is completed within five hours. Sample loading, electrophoresis, data collection, and data analysis have been automated to allow unattended operation. Because it is a capillary electrophoresis instrument, there is no need to manually pour or load gels. Benchmarking shows length of reads as long as 550 base pairs with 98% accuracy, depending on template and primer quality. The 3700 is an ideal platform for sequencing PCR products less than 550 base pairs in length. It also works well for single stranded and double stranded plasmid DNA templates. The length of read appears to be slightly less than that obtained on ABI 377 and 373 instruments.
